Using genetically encoded calcium sensors, we define how zaprinast functions to activate T. gondii egress from infected host cells. We use this phenomenon to identify compounds that enhance or block calcium signaling in the parasite, describing new inhibitors of T. gondii cGMP-dependent protein kinase and a novel compound that releases calcium from intracellular stores.
